Primary outcomes
Number of correct responses, frequency of speech errors, and the content and frequency of circumlocutory responses produced during the SALA picture-naming tasks. These outcomes are assessed during the picture-naming tasks at the study visit.
Key secondary outcomes
Differences in the concepts expressed in circumlocutory responses according to target-word category, and differences in the frequency and conceptual content of circumlocutions according to age, sex, and educational background.

Participants undergo a single study session consisting of a questionnaire on basic demographic information, the Japanese version of the Mini-Mental State Examination (MMSE-J), the Naming I subtest of the Sophia Analysis of Language in Aphasia (SALA), and a study-specific picture-naming task (Naming II) developed by the research team using selected target words. The MMSE-J takes approximately 5 minutes, and each naming task takes approximately 10-15 minutes. During the naming tasks, participants are asked to name pictures presented one at a time on a computer screen, and their verbal responses are audio-recorded.

Key inclusion criteria
Adults aged 20-89 years.
Individuals who provide written informed consent to participate after receiving an explanation of the study.
Individuals with a score of 24 or higher on the Japanese version of the Mini-Mental State Examination (MMSE-J).
Key exclusion criteria
Individuals with a score below 24 on the Japanese version of the Mini-Mental State Examination (MMSE-J).

Results
A total of 140 participants were enrolled, and 138 were analysed after excluding two participants with MMSE-J scores below 24. Mean naming accuracy was 89.7%, and the mean speech-error rate was 9.2%. Semantic paraphasias were the most frequent error type, and naming performance differed across age groups. Circumlocutions were also observed in healthy adults and could be classified according to several semantic characteristics. Parts of the findings have been presented or published previously.

Baseline Characteristics
The final analysis included 138 participants: 64 males and 74 females. By age group, 26 participants were aged 20-34 years, 61 were aged 35-64 years, 25 were aged 65-74 years, and 26 were aged 75 years or older. The mean duration of education was 13.9 +/- 1.7 years.
Participant flow
A total of 140 healthy adults were enrolled. All participants underwent the MMSE-J, and two participants with scores below 24 were excluded from the analysis. A total of 138 participants were included in the final analysis.
Adverse events
No adverse events related to the study were observed.
Outcome measures
In the SALA Naming I task, mean naming accuracy was 89.7 +/- 8.1%, and the mean participant-level speech-error rate was 9.2 +/- 5.7%. A total of 1,191 speech-error tokens were identified among 13,077 naming responses, with Semantic paraphasias being the most frequent type (782 errors, 65.7%). Naming accuracy differed significantly across age groups, as did speech-error rate. Coordinate and Superordinate responses were frequent among semantically related errors, whereas Substitutions were the most frequent pattern among phonologically related errors.
