MRI評価：
各実験の10時間前から絶食する。初日は上腕部にグルコースセンサーを装着し、安静時のベースラインを15分間測定した後、75 gブドウ糖溶液を摂取する。その後2時間安静を保ち、グルコース応答を記録する。2日目および3日目は、ブドウ糖摂取条件および炭酸水摂取条件で、脳または臓器の機能的MRIを行う。T2*強調画像を75分間撮像し、撮像開始15分後にブドウ糖溶液または炭酸水を摂取した後、さらに60分間撮像を継続する。
磁気刺激評価：
参加者は、必要に応じて75 gブドウ糖溶液の摂取前後に経皮的磁気刺激を受け、脊髄・末梢神経機能を評価する。皮膚表面電位、筋電図、脳波および刺激により誘発される四肢反射を記録し、四肢運動をモーションキャプチャで測定する。

両評価では、ブドウ糖摂取前後の糖代謝関連の自律神経・臓器応答を評価する。上腕部にグルコースセンサーを装着し、糖代謝関連項目を測定するため採血を行う。同日に複数回採血する場合は、必要に応じて生理食塩水の静脈路を使用する。採取した血液から自己血糖測定器でも血糖値を測定する。採血量は1回の実験につき1日20 mL以下とし、同一参加者への採血は週2回以内とする。

MRI assessment:
Participants fast for 10 hours before each session. On Day 1, a glucose sensor is attached to the upper arm. After a 15-minute resting baseline, participants ingest 75 g of glucose and remain at rest for 2 hours while glucose responses are recorded. On Days 2 and 3, functional MRI of the brain or organs is performed under glucose and carbonated water control conditions. T2*-weighted images are acquired for 75 minutes. Fifteen minutes after scanning begins, participants ingest glucose solution or carbonated water, and scanning continues for 60 minutes.

Magnetic stimulation assessment:
Participants may undergo transcutaneous magnetic stimulation before and after ingestion of 75 g of glucose to assess spinal and peripheral nerve function. Skin surface potentials, electromyographic and electroencephalographic activity, and stimulation-evoked limb reflexes are recorded. Limb movements are also recorded by motion capture.

In both assessments, glucose-related autonomic and organ responses are evaluated before and after glucose ingestion. A glucose sensor is attached to the upper arm, and blood samples are collected for glucose metabolism-related measurements. If repeated sampling is required on the same day, an intravenous line with normal saline may be used. Blood glucose is also measured with a self-monitoring meter. The total blood volume is limited to 20 mL per experiment per day, and sampling is performed no more than twice per week per participant.

BMIが18.5未満あるいは30以上
収縮期血圧が140mmHg以上
拡張期血圧が90mmHg以上
習慣的な喫煙（毎日1本以上の喫煙）
糖尿病の典型的症状（異常に喉が乾く・頻尿あるいは多尿・疲れやすい）がある
これまでに健診などで糖尿病の診断を受けたことがある

＜MRI計測用除外基準＞
閉所恐怖症や体内金属のある者などMRI禁忌事項に該当する者
参加者選定用アンケートでMRI禁忌事項に該当する者
MRI測定実施日のチェックリストでMRI禁忌事項に該当する者
強度の磁場酔いおよび脳形状に異常があれば研究開始後に除外

＜経皮的磁気刺激用除外基準＞
てんかんの既往のある方、心臓ペースメーカーを装着している方、頭部に金属を埋設した方、及び妊娠が疑われる方。

<Common exclusion criteria>
Younger than 18 years or 65 years or older
Body mass index (BMI) below 18.5 or 30.0 or higher
Systolic blood pressure of 140 mmHg or higher
Diastolic blood pressure of 90 mmHg or higher
Regular smoking, defined as smoking at least one cigarette per day
Presence of typical symptoms of diabetes mellitus, including excessive thirst, frequent urination or polyuria, and fatigue
A previous diagnosis of diabetes mellitus during a health checkup or by a physician

<MRI>
Individuals with contraindications to MRI, such as claustrophobia or metallic implants or devices in the body
Individuals identified as having MRI contraindications based on the participant screening questionnaire
Individuals identified as having MRI contraindications based on the checklist completed on the day of MRI scanning
Individuals who experience severe magnetic-field-induced vertigo or in whom structural brain abnormalities are detected after study initiation

<Transcutaneous magnetic stimulation>
Individuals with a history of epilepsy, a cardiac pacemaker, metal implants in the head, or suspected pregnancy.
