日本語
医学的適応または社会的適応による卵子の凍結保存の施行件数は増加傾向にある。一方で、胚を凍結融解することで発生能低下や細胞骨格の分布変化が引き起こされることが報告されており、凍結融解技術の改良が求められる。そこで我々は融解後卵子/胚の脂質代謝に注目した。マウスの卵子/胚を用いた実験において、融解後に胚発生のエネルギー源となる脂肪の細胞内含有量が減少すること、また融解液への脂肪酸の添加がその後の発生能およびエネルギー代謝活性を改善することを明らかとした。さらにウシ初期胚を脂肪酸添加融解液により融解することで胚盤胞への発生率が向上した。また研究への使用に同意が得られたヒト胚を用いた試験においても、脂肪酸添加融解液の使用により胚の形態学的評価が向上することが示された。そこで本検討ではヒト凍結卵子の融解に脂肪酸添加融解液を使用することで着床前胚発生が改善されるか否かを検討する。

Vitrified metaphase II stage oocytes exhibit a diminished ability to develop into blastocysts and live births. Previous studies have shown a reduction in intracellular lipid content as one of the factors associated with reduced developmental competence of oocytes after vitrification as the intracellular lipid content of oocytes is affected by vitrification. The fatty acids (FAs) derived from break down of lipids are primarily transferred to the mitochondria, where it plays a crucial role in cellular metabolism. A recent study reported that FA supplementation in the warming solutions improved the developmental competence of the vitrified-warmed mouse oocytes by activating the beta-oxidation pathway. Furthermore, FA supplementation enhanced the developmental rate to the blastocyst stage and improved morphological characteristics of bovine and human embryos vitrified at the cleavage stage. In the present study, we examine whether FA supplementation in warming media influences the developmental competence of human oocytes after warming.
